I am doing project on the Th17-dependent neutrophilic asthma based on the papers published by Dr. Gregory S. Whitehead and Dr. Donald N. Cook as you can see in the attached files. Briefly, The BALB/c mice were sensitized intranasally with 0.1ug LPS and 100ug OVA in 50ul saline solution on Day0 and Day7, and challenged on Day14 for 1h with 1% OVA and sacrificed 24h later. Now my questions are as follows: The levels for IL-17 in the BALF is only about 30ng/mL, which is much lower than the results published by Dr. Gregory S. Whitehead et al., I am really confused by this problem. Thank you very much!
I did not read all details of your references but it seems that Whitehead and Cook mostly used C57BL/6 mice for their esperiments. The different level of IL-17 might thus depend on the fact that Balb/c mice in general are weaker responder to LPS.
Thank you very much, Dr. Maestroni. It may be true that the strains of mice play an important role in the level of IL-17, as Whitehead and Cook mentioned in their responses to my E-mail. I plan to compare the models by using BALB/c and C57 at the same time.
i guess LPS dose is too low to induce neutrophilic asthma, rather such dose might induce eosinophilic one.
