So, I am unable to find a direct answer but essentially I want to use an ELISA protocol to measure the amount of ubiquitylated histone H2A in my mutant cells. I was wondering if I could get a kit that included capture antibodies against histone H2A then a detector antibody against Ub-H2a ? I ask because I have not had much luck finding any sandwich ELISA kits with anti Ub-H2a.
You will not find such a kit unfortunately as the measurements of labelled antibodies will be inaccurate since most ELISA are colorimetric/Chemiluminescence. Your best bet is Western Blotting with anti-Ubi H2A and there are many specific to ubi residue, followed by intensity measurements to get relative quantitation. Or you can make your own Elisa using available MAB of choice with custom made ELISA kits available for example abcam (https://www.abcam.com/simplestep-elisareg-custom-elisa-kit-ab270552.html) and LSBio (https://www.lsbio.com/products/elisakits/custom)
I think you use the detectable antibody of ubiquitylated histone H2A and then 2nd antibody conjugated with a molecule to be reacted by specific wavelength.
