For quantitative cellular uptake of nanoparticles on HCT116 and A549 cells, after removing the nanoparticles treatment from the cells, should I detach the cells using trypsin or I can directly lyse the cells with MeOH/acetonitrile before injecting into LCMS for drug quantitation?

Yes, you have to detach and centrifuge the cells.
These are my suggestions:
1. Detach, wash and centrifuge the cells.
2. Collect and lyse the cells pellet.
3. Sonicate to extract the drug out. Be mind the condition (ambient or ice bath) during sonication (depends on your drugs).
4. Centrifuge and analyze the supernatant using LC-MS (appropriate mobile phase of your drug).
Others may have a good method.
