Dear colleagues,

I am setting up an in vitro stimulation of mouse splenic B cells with LPS, CpG and IFNb. The read-out will be qPCR for cytokines and IFN-regulated genes.

For B cells purification I am considering to use positive selection with CD19 MicroBeads or negative selection with B cells isolation kit (both from Miltenyi). I am wondering how positive or negative selection for B cells can affect the stimulation and the results? Does anyone have experience with this?

Finally, what if I want to stimulate the BCR receptor with IgM for example always on splenic mouse B cells. Can the positive selection CD19+ MicroBeads interfere with the stimulation?
