17 February 2017 2 929 Report

Hi all!

Our lab is trying to get all of our rules and protocols straightened out and one that we aren't sure about is the best way to store and take out cryosectioned slides.  We currently add desiccant to the box to help with moisture but the question that has come up is do you have to wait until the box is at room temperature to open it?  So for instance, we want 2 or 3 slides from a 100 slide box stored in the -80, can we just grab the slides quickly or do we wait for the whole box to come to room temp before opening?  Typically we just grab the slides as quickly as possible and return the box to the -80, however we want to know the best way to preserve the tissue on the slides.

Our tissue is typically fixed with 4% PFA.

Would love to know what you guys do in your lab.

Thank you so much!
