I would like to air dry the cell suspension on coated or non-coated glass slide but the salt crystals of PBS ruined the fluorescent image. I have to count all the cells in the suspension so I cannot smear the little volume of suspension on the slide. Do you have any suggestion or protocol for air drying?

I found this protocol (https://www.rndsystems.com/resources/protocols/protocol-preparation-and-fluorescent-icc-staining-non-adherent-cells), they resuspend the cells in the DI. After fixation with formaldehyde, can I wash the cells with DI without lyse them?
