I work with the protein CWC2 which contains a zinc finger. My question is during the process of purification is zinc removed from the protein? If so, how should I go about re-adding it?
The effect of purification process on zinc removal from the protein is depending on the purification method and purification conditions . for example if you used EDTA or citric acid in separation buffer or used very low pH buffers that will remove zinc from protein.
To add to the answer of Jasim Al-saadi, unless you are sure that the protein survives Zn-removal, I'd be careful with any treatment that removes this cofactor. In some proteins, cofactors are added co-translationally, they get denatured irreversibly if the cofactor is removed.
