Hi, anybody knows how fast a bone marrow sample for Fish or conventional cytogenetics taken from bone marrow and placed on McCoy medium needs to be processed?
I work 5 hours away from the Research facility that does our cytogenetics and the admin personnel tell me it needs to be at the facility the morning after it was taken.
Is that so? even in a culture medium? can there it be an interval of 36 hours between taking the sample an it reaching the facility?

Faster the better, or else quality of metaphase, yield, etc. can be affected. Also, the downstream steps of colchicine addition etc. are best avoided in odd hours hence the planting time or initiation of culture time has to be back engineered. Other constraints can be addressed but optimum quality is a prime concern.In case of bone marrow, especially in leukemia cases, the number and proliferation rate of dividing cells which we want to trap at the metaphase are not uniform, vary a lot, and hence necessary pre-processing has to be done, like cell count etc. to start the culture. If left for a long time, in the presence of media, the cell proliferation will continue and more asynchronous mass of cells will be there at the time of culture setting. Keeping the sample with media at low temp. is one of the approaches for getting synchronized culture, however, our experience was for about 5 hours. As per the logistics, one has to device best suited protocol. Hope this helps.
