Please advise/help me on the preparation of chitosan hydrogels. Currently my procedure looks like this:
Step 1. Dissolve chitosan (1g) in 20 ml of 2% acetic acid.
Step 2. I add the previously prepared solution from step 1 to a 10 molar solution of NaOH. I am forming the target disc shape, but the crosslinking is weak.
Step 3. I dip the disc from step 2 into a 1.5 mole solution of CaCl2 for better cross-linking.
Unfortunately, I do not obtain a rigid hydrogel. The material is disintegrating.
Can more experienced researcher suggest what is wrong with this procedure or how to improve it to get a rigid chitosan hydrogel.

For preparation of chitosan hydrogel, glutaraldehyde can be used. I am not sure but to obtain rigid hydrogel, you can use 0.25% to 1.0% (v/v) glutaraldehyde solution mix in chitosan solution.
Effectivement, il faut dissoudre le chitosane dans une solution d'acide faible, comme l'acide acétique ou bien l'acide lactique.
Une fois dissoutes, les molécules de chitosane portent une charge positive.
Pour obtenir un hydrogel rigide, il faut faire réagir ces molécules chargées positivement avec une solution d'un polymère chargé négativement, comme l'alginate ou le polyphosphate.
