I have problems growing Arabidopsis on MS plates. The plants are small, reddish and only small fractions of plants germinate. I have tried it by myself (previously a student did it) and it's better (at least they do not seem to be so reddish), but the germination rate is still far from 100% even for WT on plates without antibiotic.
The problems could be of two origins:
1: Medium - I use full MS (commercial) without addition of vitamins.
2: Handling of seeds - here I see maybe the biggest possibility for problems. I know it's crucial the keep the seeds in EtOH only for short time during sterilization. Can it decrease the germination rate by 20-30% for elongated EtOH wash?
How are the seeds best stored? I keep them in eppies at RT (one of our post-docs keeps them at RT as well, and he seems to be quite experienced with Arabidopsis) and in the dark. Is it important to keep them like that for some time before germinating them, or can one collect them and immediately germinate (based on the number of small plants that manage to grow in the pots after giving seeds, I'd say it doesn't really matter)?
What else could be the problem?
