I have a problem with so high absorbance in crystal violet method. I mixed monomeric quaternary ammonium compounds, biofilm and crystal violet. In higher concentration of QACs I have so high abnormal absorbance. So maybe you have any idea what goes wrong?
The earlier response by Andrew Spiers to a similar question refers: Briefly, "you should make sure that you have as many replicate wells just filled with growth medium (negative controls) as you have replicates fro each of your strains (treatments), and process them in the same way. When you measure the absorbance (not optical density) to determine the levels of eluted crystal violet, you can compare the negative controls with the other treatments".
