In lentiviral plasmids, the WPRE is usually placed near the 3'LTR, in the 3'UTR of the transgene.

My question is; can WPRE be placed in the antisense direction, so near the 5'LTR, in-line with an antisense transgene and antisense promoter like this;

5'LTR-antisense[polyA-WPRE-gene1-inducible promoter]-3'LTR

My question relates to the "ideal" placement of genetic elements for a dual-promoter lenti plasmid, which is not trivial when taking into account place restriction and interference of promoters with each other and with the 5'LTR.

Would something like this work;

5'LTR-antisense[polyA-WPRE-gene1-inducible promoter1]-insulator-constitutive promoter2-gene2-WPRE-3'LTR
