I isolated the on PDA in leaf litter samples. at the moment i cant see any clear spores, or conidiophore under the microscope. I am culturing them on water agaar to induce sporulation
I suggest for best result, re-inoculate them on PDA for 4-5 days maximum, then see the conidiophores under microscope using sticky-tape technique with one drop of methyl blue.
The yellow color colonies may be of Aspergillus candidus or A. ochraceus and the red colored colonies of Fusarium graminearum, F. culmorum. Please observe the shape and sizes of conidia under microscope and consult Manual of Aspergilii and Nelson et.al., 1983, "Fusarium species: An illustrated manual for identification" to confirm the species.
To make taxonomic identification to genus (at least), is necessary to observe micromorphological aspects such as the characteristics of the spores, fruiting structures and structures of resistance.
If any of the isolated not sporulate you can try to cultivate on Synthetic Nutrient Agar and exposed to cycles of light and darkness. Keep in mind that there are also sterile mycelia.
