I am trying to isolate antioxidant compounds from red algae so can anyone share with me any protocols or journals related to glass column chromatography?
I'm not sure whether these are "standard" procedures, but they do work for me.
I take extracts and run them on C18 (non-aqueous reverse phase for non-polar antioxidants) or strong anion exchange columns followed by Sephadex LH-20 and C18 (for phenolic compounds). Polyamide resin would also work for the phenolic compounds as a clean-up step. I've isolated tocopherols from oils with a diol column.
Bioautography detection in thin-layer chromatography is suitable method to detection antioxidant compounds from red algae and based on this you can isolate antioxidant compounds. Please see articles regarding bioautography detection in thin-layer chromatography.
