If I understand well the question you intend to extract plasmid from yeast. So what you have to do is to grow your yeast strain in selective media, harvest cells  and run the following protocol which relies on the use of miniprep kit reagents:

In your case yeast has to be cultured in CSM lacking uracil. And selection of bacterial transformants will be made on ampicillin containing media. Then classical miniprep can be performed to produce substantial amount of the plasmid.
