I am doing a research on the effect of 4 polyphenols on kainic acid treated glioblastoma cells. I used a multiplate reader machine to measure cellular proliferation of the 4 polyphenols at different concentrations. Statistically analyzes should be done to investigate the effect of each polyphenol. From journals read the measurement for each polyphenol starts from a single point thereby making determination of IC 50 possible. I cannot seem to get that with my data. Guess its something I need to do with my control values in relation to others.
The user manual for the plate reader should provide guidance if nothing else. The manual should describe the different analysis options and good manuals may provide examples where the different methods are used. It could be an issue if you did not have (or did not tell the machine that there were) control wells. The options that you want may not be displayed without the proper controls.
Since the machine does the analysis for you, we would need the make and model of the machine to provide other help. You might also contact the manufacturer of the machine. They can be helpful, and they should know how to use their equipment.
