I want to add 1 ug or (1000ng ) of Plasmid DNA in a reaction mixture and add water for a final volume of 50 ul. my plasmid DNA a concentration of 68.47 ng/ul in which I have total DNA volume 25ul. How much ul should i have been used into my mixture? dear any one help me. Thanks in advance

If your DNA concentrations is 68.57 ng/uL, and you need a 1000 ng (1 ug), you should use 14.604936 uL of this solution.

Concentration(start) x Volume(start) = Concentration(final) x Volume(final)
This equation is commonly abbreviated as: C1V1 = C2V2
Note: The diluted concentration must be less than the stock concentration
1000/68.47= 14.60 ul of plasmid DNA
50-14.60 = 35.4 ul of H2O
(N1V1=N2V2)
Mesele Tilahun 14.60 uL of plasmid is correct. But, you should not add 35.4 ul of H2O as suggested in Bhavesh's answer. Because you are making a reaction tube which has other components to add.
14.60 uL (plasmid) + other components + water = 50 uL
Yuan-Yeu Yau , yes I now water is the rest component fill unto 50ul. I use 14.6ul pDNA+ 10XEZbuffer + enzyme and the rest volume is water. my target question is to know how I calculate my DNA conc. into ul? .thank you your thinking for considering of other components.
