I have prepared plant extract of 0.1 mg in 1ml of petroleum ether..(stock solution)
Now I want to do antibiotic susceptibility test in different concentrations around ( 25-100 microlitre)..if I take 25 microlitre from stock what will be the concentration ? Or shall I prepare different concentrations like 25;50;75;100 ?
Whatever volume you take from stock solution, the concentration would be the same. You need to dilute stock to get lower concentration.
Hi - 0.1 mg in 1 ml would be 100 microgram (mcg) in 1000 microliter (mcl), so if you take 25 mcl of this stock solution, it would give you 4 mcg of the extract. You did not mention what model/system you are using for antibiotic susceptibility test (i.e., disc diffusion, microdilution, etc.) but normally you want to keep the volume constant for all concentrations tested. So preparing different concentrations (from a single stock solution using serial dilution technique) is recommended as opposed to taking different volumes of the same stock solution. You might also want to check if it is possible to use other solvents for the preparation, as petroleum ether might be a problem in some test systems.
Ok. thank u sir...I am willing to do disc diffusion technique..but I have more research papers with well diffusion...so I will try both..whichever results are better that I will keep for further work
