I have a question regarding PKH26 labeling.

I want to analyze the ability of cancer cells to uptake EVs into their cytoplasm. What I did was I stained EVs with PKH26 as described on the manufacturer's protocols (Sigma) then co-cultured them with cancer cells. I also stained the cells with DAPI at the end of co-cultured period to stain the nucleus and to ensure that internalized EVs are located in the cytoplasm.

Unfortunately, under the fluoresence microscope, I found several purplish dots in cancer cells' nucleus. I thought that the purplish dots may come from the PKH26. However, I did not expect that PKH26 will stain the nucleus as well.

Do you have any opinion or suggestion regarding this issue?
