I have a question regarding PKH26 labeling.
I want to analyze the ability of cancer cells to uptake EVs into their cytoplasm. What I did was I stained EVs with PKH26 as described on the manufacturer's protocols (Sigma) then co-cultured them with cancer cells. I also stained the cells with DAPI at the end of co-cultured period to stain the nucleus and to ensure that internalized EVs are located in the cytoplasm.
Unfortunately, under the fluoresence microscope, I found several purplish dots in cancer cells' nucleus. I thought that the purplish dots may come from the PKH26. However, I did not expect that PKH26 will stain the nucleus as well.
Do you have any opinion or suggestion regarding this issue?

it looks like you got your experiment right. There's seems to be flow of PKH26 from the vesicles into the membranes of cancer cells. So you got an excellent conclusion. Net flow of materials.
