Using just PBS isn't enough to disrupt cell membranes and effectively lyse cells. You will have to add an non-ionic detergent like Triton X-100 at a very low concentration (0,1-0,5%). Don't forget to add protease inhibitors and do the whole process using freshly prepared and ice-cold solutions. Make sure the cells are incubated with the lysis buffer for at least half an hour. Gentle sonication might also be useful.
