I am looking at papain induced airway inflammation in LysM cre BMAL1 mice. I have used following dose.

Dose: (Animals were on Isoflurane anesthesia)

30 µg in 40µl/ animal, intranasal challenge

( Kamijo et al., DOI: https://doi.org/10.4049/jimmunol.1201212 )

However, the mice are bleeding through nose and mouth immediately after the challenge. When i dissected the lungs, I see lung hemorrhage. Has anyone encountered similar incidence? Is this because of the type of papain (# 76220-25G, SIGMA) I have used? I have read articles where people have used 80 micro gram/40 micro liters of papain in mice .

Any kind of help is highly appreciated.

Thanks

Amruta
