@David Pires thank you for your kind advice. Actually we are using stray method and just needed some kind of adaptation/optimization because we are applying this same method for other soil samples. We also thought about mixing half the forest soil sample with sterile soil ending with two suspensions.

You can use the Cobb method (decantation and sieving). However, the Cobb method requires more time to process soil (per sample unit).
