I have iPSC and primary fibroblast lines that constitutively express dCas9, and I am planning to screen RNA guides produced by IVT in these cells. Since the cells already express dCas9, I only need to introduce the gRNAs (both individually and in combination) into the cells.

  • What is the best transfection protocol to use in this scenario? Should I use lipofectamine or electroporation?
  • What concentrations of gRNAs should I test for optimal results?
  • Any advice or recommendations would be greatly appreciated!
