Well, I want to express protein using OmpA on bacteria surface(outer membrane).

so I extracted e.coli DNA from ETEC and then PCR to get OmpA.

originally, I expected that the sequence of OmpA is 1a.a. to 159a.a. containing signal peptide.( you can easily find this sequence in some papers if u know OmpA as surface anchor motif)  

But it was different. When I check it by sequencing, It has quite different amino acids on hydrophilic region. so total OmpA size(about 1aa-185aa) is longer than 1aa-159aa OmpA expected. but there are no difference on hydrophobic region.

 I know that the hydrophobic region of OmA is important for their fuction. so I don't think binding ability to outer membrane of 1aa-185aa OmpA will not be influenced by difference of hydrophilic region. But I'm not sure and I have not been able to find some papers about this OmpA for surface display. (1aa-185aa OmpA sequence exists in some E.coli for 100% match)

I want some advise about this OmpA(1aa-185aa) for surface display. Anybody who knows about this OmpA, please comment. related papers are also welcome.
