In the process of determining oil content of a doughnut-like snack, acid hydrolysis was carried out, before Soxhlet extraction with ether. the extract came out black. Is this normal? or which extraction method would you recommend? Thanks.
You can determine the oil content without going to the step of acid hydrolysis. While i do not know why you attempted acid hydrolysis, in case, you want to do that, use another portion of sample for soxhlet extraction to determine the oil content.
Thanks for your responses. I used acid hydrolysis because of a method i was following. I had to discard it later. Now i am just extracting the dried sample using Soxhlet.
Acid hydrolysis could be done for the release of bound lipids as in the determination of lipids from milk or other diary products by centrifugation. If strong sulphuric is used the organic mater is destroyed and turned black.
For the bakery products and biscuits Soxhlet method is used with good results.
