I have developed the kit successfully until half strip.

But I could not replicate the same results in full strip development.

I have changed sample and conjugate components with a famous brand.

Still I am not getting results.

I am using same polyclonal ab for both conj and test zone. I am using 1 mg/ml test zone antibody conc and 25 nm Gold nanoparticles.

Sometimes I get ghost zones in the test.

Can any one suggest a better blocking / running buffer and processing steps for sample and conj pads to replicate the testzone results in the complete strip?
