Your NCE is usually stored in DMSO at a high concentration (>100mg/mL); add incremental amounts (microliters) of the DMSO solution to water (maybe at a pH of your choice using a buffer, record the ionic strength). Do not exceed 2% of DMSO in the final solution because it will give false solubility due to the DNSO partition. Determine concentration by HPLC using a calibration curve or by turbidimetric method (record UV/VIS at a wavelength where there is no absorbance of your compound. when the concentration exceeds the solubility, it will crash out, thus turbid solution and increase in absorbance).
These leads are not stored in DMSO yet but vials within a desiccator. That's why I want to run a solubility study to determine their aqueous solubility behavior and in other solvents. so I am looking for a study protocol to follow.
