In therapeutic analysis for in vitro cell based assays, solubility of the chemical molecules is the most important problem. For water insoluble compounds we generally prefer DMSO (Dimethyl Sulfoxide) but it has some limitation like, many molecules are sparingly soluble and DMSO has toxicity at higher than 1-5%. Thus kindly suggest any non-toxic solvent for cell based analyses.

We would use DMSO as a standard solvent for new compounds, never more than 2 %. You may consider that a molecule which you have to dissolve in more than 5 % of DMSO can have limited therapeutic potential. So don't focus so much on increasing your solubility using solvents, consider to make the compounds more soluble in itself (pH changes, if possible, formulation as salt, amorphous formulation)..
