Hi,

I would like to know if somebody is currently using expression plasmids of RIPK1, RIPK3 and MLKL? And if so, anyone has some mutants that leads to constitutively kinase activity of those, thus triggering necroptosis in cells such as L929, HT-29, Jurkat FADD (-/-), BV2, etc?

The main purpose of those plasmids for me would be to further explore the potential target of some new necroptosis inhibitors.

I would like to transiently transfect cells with each plasmid, thus inducing a significant amount of cell death, and than co-incubate the compounds to further understand whether they target downstream or upstream of RIPK1, RIPK3 or MLKL.
