-The development of NEW treatment strategies for rheumatoid arthritis (RA) remains a priority.
-Despite recent advances in RA treatment attributed to biologics, only a minority of patients achieves and maintains remission. Treatment of RA is complicated by the development of tolerance over time or failure of patients to respond to currently available therapies.
-Nanotherapy in RA is receiving increasing attention with potential advantages.
-Nanocarriers (submicron transport particles) are designed to deliver the drug at the site of inflammation — the synovium — thereby maximizing its therapeutic effect and avoiding unwanted systemic adverse effects. This Targeted Drug Delivery approach minimizes the amount of drug required to control arthritis and increase local bioavailability.
-A “redesign” of already effective rheumatologic medications into nanoformulations may confer greater specificity, therapeutic effect and a more amenable safety profile.
-Nanoencapsulated NSAIDs, liposomal and polymeric preparations of glucocorticoids and nanosystems that directly inhibit angiogenesis are examples of nanotherapies that have been tested in experimental models of inflammatory arthritis.
-Despite the promising findings observed to date, further development and subsequent integration of nanotherapy in the management of RA remains hampered by the lack of efficacy and toxicity studies in humans.
