The contamination that I see in my host cell flasks and thus also my parasite cultures are very much different from the usual bacteria, mycoplasma or fungal contaminations encountered in routine cuturing of T. gondii in a HFF background. What I see in my flasks are semi transparent greyish round bodies (perfectly circular) that mostly float, but also attach onto the cell surface and seem to invade and form vacuoles too. Since the past three weeks I have faced this problem. I tried throwing away all my host cells and making new flasks, new medium with best efforts to avoid contamination risks, but I still have the trouble.
Kannan, same problem I am also facing but for me only one cell line I had that problem. My suggestion is, If the vials are frozen down from the same mother culture then you can't get rid of that cos sometimes these circular structures stay inside the cell and you can't see the media turbidity. If it is frozen down at that time these contamination will show its aggressiveness while thawing. So, go for very earlier passage numbers if possible. or otherwise check whether this contamination is present in all your other cell line. Hope you have checked your media and FBS separately for contamination (I checked for mine but they are free of contamination) I seen these circular bodies only with cell lines and the great thing is unlike other bac or fungal contamination media turbidity you can see after passage or two to three days later.
