I have HEP-G2 in culture with DMEM (4.5 g/l Glucose, +Glutamine, -Pyruvate). I exchanged the media every 2-3 d. Unfortunately, the cells are not growing as intended and I couldn't passage them since thawing two weeks ago. What can I do?

Hi,
HepG2 cells need higher cell density to grow and a lower glucose content (~ 1g/l). Some companies adapted the cells to RPMI medium instead of DMEM.

Hi Robert,
what exactly do you mean when you write "cells are not growing as intended"? HepG2 cells exhibit a special morphology and tend to form clusters instead of a clear monolayer. So you will actually never get a dense cell morphology, no matter how long you culture them.
If you are aiming for a more even distribution of your HerpG2 cells try to coat the surface of your cell culture dishes with polyethyleneimine first - that helps a lot and cells distribute evenly on the surface!

Thanks for your answers. The cells weren't growing at all, after two weeks I saw them forming clusters. But they grew very slowly. Hope changing the medium helped.
