Hello everyone,

I’m testing bacterial extracts on cancer cells. MTT assay after 24 hours (repeated multiple times) showed a significant reduction in viability at the IC50 concentration, along with visible morphological changes (cell shrinkage and detachment).

However, when I performed Annexin V/FITC apoptosis assay at the same IC50 concentration and duration, the results showed ~87% viable cells and ~10% necrosis, which contradicts the MTT results.

I understand that MTT is a metabolic assay and doesn’t always reflect actual cell death. Could the cells be metabolically arrested but not dead? What could explain this contradiction, and how should I troubleshoot further?

I would appreciate any suggestions and relevant reference papers.
