We were trying to establish this TRAMP-C1 syngeneic tumor model in our lab. This cell line was obtained from ATCC, and we tried subcutaneous injection into B6 mice (8-12 weeks). However, what we encounter is very low tumor rate...we tried a few times, but the result is disappointing....any suggestion for successful result?
Based on literature, we know it is a slow growing tumor....it may take 45-50 days...however, we wait almost 2-3 months...but nothing comes out.
Good question. I don't think I can help you, as my lab does not have experience with this model.

I am afraid that the same thing is happening to us. We have purchased TRAMP-C1 and TRAMP-C2 from the ATCC and neither of them grew in C57Bl/6 mice. Even worse they did not grow in NOD/SCID mice !!! We complained but they refuse to do anything about it!!! You can try injecting the tumour with materiel and use older mice that is all I can recommend.

We had a similar problem with A20 cells giving variable results. We cloned them, looked for 4-1BBL expression (endogenous) and then found those without 4-1BBL expression formed tumours consistently. These don’t seem to be working at all but you could check their MHC or for other markers that they have the expected features of TRAMP-C cells
Stephanie E.B McArdle We may find a way to increase tumor rate of this particular cell line. We inoculated cells into nude mice and waited for the tumors to grow. We isolate tumors and dissociated into single cells. Then, we injected cells back to nude mice for repeating 2-3 times. Later, we found, after this selection cycle, the tumor rate was much better than we just received from ATCC. It was a tedious work. But, at least, we finally made it work better.
Thank you. Since my last post Dr. Matteo Bellone gave us some of his TRAMP-C1 cells and we were able to grow these successfully in both C57Bl/6n mice and NOD/SCID. I have also taken some of the tumour that grew onto the NOD/SCID and re-derived them in vitro. I will now be testing them to see whether these are more aggressive.
