I am trying electroporation for mouse İPSC. However my efficiency maximum around 15%. I would like to change parameters or I can use different transfection method like PEI. Can somebody share with me experience about mouse iPSC transfection.
In my experience, the most important thing during transfection of iPSCs is to catch the moment when they are in exponential growth (when there are many-many dividing cells and cells look the liveliest). If you try to transfect them when they are in other phases, it's extremely difficult. And also, 15% is not that bad in iPSCs.
To better catch the exponential "high point" you can try transfecting them by a lipid-based method, like lipofectamine or FuGene. It may work better in some cases, possibly because in some cell lines when you dissociate cells from plate surface, they stop dividing and it takes time for them to gain momentum again.
