I have come across an IVD quantitative assay for C-peptide where both anti analyte biotin-conjugated antibody which binds to Strep coated wells and HRP conjugated anti-analyte antibody was also raised in murine. Wouldn't these antibodies cross-react and generated false positive or is it a common practice of using capture and detection mAbs raised in the same species.
If the two antibodies are monoclonals recognizing different, non-overlapping epitopes, and you directly use a HRP-conjugated anti-analyte antibody, rather than a secondary anti-mouse IG antiserum to detect the bound analyte, this is no problem.
If these are both raised in mice, then there is not a concern the capture and detection antibodies will cross-react. One mouse antibody would not bind to another mouse antibody. The only concern would be if the monoclonal antibodies bind to the same epitope of the analyte, because then they would compete and you would have poor signal.
