Hi!

I am doing some microinjection of cells, but sometimes, they get more damaged than usual. My question is: For the fast reparing of membrane integrity after permeabilization experiments, a solution with 2 or 3 mM calcium is added, so that the cell does not get that much damage during the proceedure. For what I understand, the higher amount of calcium should help to seal the membrane in this protocol. Using the same principle, should I add extra calcium to the microinjection media, so that when the needle comes out, the cell repairs itself faster? or by doing so do I risk activating signalling effects dependent on calcium fluctuations?
