I´m trying to detect a specific histone methylation mark (H3K9me2). Which is the best way to detect this type of modifications? I have tried WB analysis but I don´t have consistent results. I also tried imunofluorescence. Do you recomend any other method? A more reliable one?

Which antibody have you used for WB analysis? In my case the mouse monoclonal antibody from abcam (ab1220) gave consistent results.
However, one of the most reliable ways for histone PTM quantifications is LC-MS/MS. This is especially the case if you are looking into bulk levels of your modification. In case of specific genomic regions there are quantitative ChIP methods that you could try out (e.g. MINUTE-ChIP).
