Hi, we are working on antioxidants and routinely use these parameters with old referenced articles. We have observed the several limitations of these conventional methods and still want to use these parameters. It would be great if anyone could share protocols for these biochemical estimations with all the precautions that we need to keep in mind.

It really depends upon exactly what you want to get at from your measurements. GSH is very sensitive to experimental conditions and it's not unusual for a very significant conversion (up to 100%) to GSSG. Different ROS respond differently to assays. Even how you take your sample can have a huge impact on what your results are. A good answer to your question would probably require a book, not a few lines.
