Hi Guys,

I am trying to study the function of my gene/protein of interest in transcription and cell survival. The protein product is a transcription factor and has several isoforms due to alternative splicing and alternate transcription start sites. 

My current experiment involves knocking down (using shRNA) the gene, which is giving me a phenotype (cell death and changed morphology of cells in culture. The shRNA would presumably target all isoforms leading to a reduction in expression of each of them. On a western blot this indeed seems to be the case. I am wondering if anybody has had any success with knocking out specific isoforms of their target gene?

I would really like to do this to be able to dissect the function of particular isoforms. Any suggestions will be helpful.
