Is there a method to determine GSH and GSSG in plant material using spectrophotometry but without using GR nor NADPH?
I use the Noctor and Foyer (1998) method but it needs expensive reactives such as NADPH and GR
I am thinking of the Ellman reagent to check for acetylcholinesterase inhibitory activity of compounds. My reason is that, the Ellman reagent reacts with the thiol group of the modified acetylcholine which is acetylthiocholine iodine to produce a color yellow product,TNB,that can be detected by spectrophotometry at 412 nm.
I think the thiol group of GSH would react with the Ellman reagent alone to also produce a yellow-colored product.
Hope this helps.
HPLC maybe! But this depends on the sample matrix, concentration... and is expensive!
