I have used DMEM supplemented with 10% FBS and antibiotic/antimycotic for expanding. The reason for this as the expansion medium is that alpha-MEM contains ascorbic acid and will promote collagen matrix formation which will make passing difficult.
For differentiation I have used alpha MEM supplemented with an additional 50 ug/mL ascorbic acid, 10 mM beta-glycerophosphate and 10-8 M dexamethasone. I would not keep the beta-glycerophosphate in the culture over the long term but add it at the last few days of differentiation.
