Hi, I am planning to do some mammosphere assay for MCF10a cells. This is what I have tried doing so far -

Coat the 6 well with agarose and seed 10,000 cells/well with normal MCF10a media without serum. I am not sure if this is right. Can someone please tell me an inexpensive method of making this work ?

The MCF10a media has

DMEM/F12, 500ug Hydrocortisone, 100ug EGF, 50ug cholera toxin, 5mg insulin to the whole 500mL solution.

I am unable to find papers that has used the existing media that is mammocult or methocult and such. Has anyone found success using the existing media composition?
