I am trying to measure the negative generation by transpiration on the closed system (containing water and has isotropic nano-pore membrane), but I can’t detect the negative pressure although I can see some meniscus.
For the measurement of negative pressure, I used a micro syringe( volume is 10 uL). But still there is no change in syringe.
Are there any problems in my system, I wonder that the volume of syringe was not changed although negative pressure should be generated.

I do not exactly know your system, but measuring a pressure with a syringe is a bit tricky as you first need to overcome the static friction of the syringe.
Your system needs to be leak free. Further you need to take into account that when the syringe is moving the negative pressure is partially compensated by changing the volume. In order to see some movement you need a large volume.
