I am culturing human NK cells isolated by negative selection from a leukopak. I plan to activate them with either IL-15 or IL-2. What markers should I be looking at to determine whether or not the cells are activated?
NKG2D would be my choice. It's a surface receptor that is up-regulated after activation. It depends how long is your stimulation (5 hours or days). For a quick stimulus, CD69 is another alternative. NKp46 has also been reported to change after human NK cell activation.
The NK cell evaluating receptors verifying if they were ready were told by Gabriel Ascui, being the best option NKG2D to verify their effectiveness of activation would have to be made in vitro tests verifying the cytolytic potential of these cells in culture.
