What is the usual recommended procedure for maintenance of the PFGE electrophoresis cell during long breaks (not in use for 3 months):
1. Clear all the buffer/water from tubings, OR
2. Flush once a week with pure water!
I guess we are worried about the algae growth in the tubings, interfering with the electrophoresis process!
Drain the buffer completely, flush the electrphoresis cell at least thrice with MilliQ water (or double distilled water). After flushing, drain the distilled water completely, remove the tubings and keep the lid a little open. This is what we practice and we have used it successfully even after 2 year breaks.

in our lab we flush first with bleach and pure sterile water at a ratio of 1:10. Then with pure sterile water only.

Do you perform PFGE for outsiders (on a collaborative basis or commercial basis)?
We do perform for outsiders but it depends on the availability of time. As of now the usage of PFGE is infrequent but we do DGGE analysis routinely for microbial community structure analysis & identification.
