Hi everyone,

I am currently analyzing the effect of different treatments on the inflammatory phenotype of macrophages in vitro.

To add some protein data to the qPCR data I would love to stain my cells for CD206 and CD163 as anti-inflammatory markers and iNOS or CD86 as pro-inflammatory marker and quantify the % of positive cells to get a hint about their overall polarization after the treatment. However, the only papers I find do use Flow Cytometry for that matter (which sadly is not possible for me). I was wondering whether maybe ICC is not a suitbale way to analyze macrophage polarization in vitro, since it might not be sensible enough? Does anyone have experience with that? And if someone has used ICC successfully would mind sharing a working antibody?

Thank you very much! Best,

Laura
