these days, I am running an experiment which must have been simple, but I keep on receiving very disappointing results. The protocol looks really simple. In details, I am adding lysenin on red blood cells in order to induce hemolysis and release the hemoglobulin from the cells.  I am using the lysenin from SigmaMillipore. I received a lyophilized powder which I diluted in PBS to make a stock solution of c=25ng/μl. So, I have checked for different doses. I am adding 50ng/ml, 75ng/ml etc to 600ng/ml of lysenin to multiple samples of 300μl derived from the very same initial sample and they have 3x10^7 cells. Still no result. The samples that have lysenin are like the controls. No difference on OD absorption.
