We have prepared the bacteria derived gold nanoparticles and for its antibacterial and antioxidant applications we want exact concentration and for that purpose we trying to lyophilized them, but there is no success.
Kindly help me, how do I check the synthesized nanoparticles concentration.

Apparently, the bacteria molecules are bound to the surface of nanoparticles by a sulfide bond. Try washing the molecules with alcohol instead of lyophilization. If it does not work out, then it is necessary to oxidize them, and then lyophilize or rinse.

You can calculate the NP concentration indirectly by burning the sample and resuspending the ash for ICP measurement to determine the concentration of Au atoms. If you have a TEM image of the bacteria covered with AuNPs you can estimate the average size of the particles and consequentaly the number of Au atoms per single AuNP. Together it can give you an estimation for the NP concentration.
