Sorry I can't answer your question but we are trying to set up a similar assay with a TECAN 200 as well. How long are you reading the wells for? What kind of settings do you use for the TECAN? Any tips would be greatly appreciated.
I saw a paper somewhere that used something like 30min reads from memory.
Also, have you tried white plates? They can help enhance the signal from other luciferase assays that I have run.
